Research context and source limits

Discovery design and reported animal expression observations; independent human validation remains unresolved.

Discovery and experimental components

This study combines public myocardial transcriptomes with mouse experiments to examine disulfidptosis-related genes in ischaemic cardiomyopathy. The reported discovery set merges GSE5406 and GSE57338, comprising 152 control and 203 ischaemic samples. The animal protocol assigns 16 mice to each sham or disease group, with eight of each sex, and assesses tissue one week after coronary ligation.

Reported observations that can be separated

Results report reduced MYH9, MYL6 and NCKAP1 myocardial mRNA in both female and male disease groups. These exploratory animal observations do not establish human diagnostic performance. They do not establish that manipulating these genes changes disease, or that a disulfidptosis mechanism was causally demonstrated.

Validation and sex-specific boundaries

Independent validation remains unconfirmed: training accession GSE57338 is a SubSeries of validation accession GSE57345, and nonoverlapping sample selection is not demonstrated. Separately, Results report opposite MYH10 mRNA directions by sex, whereas Discussion describes matching patterns. The audit therefore excludes the claimed independent classifier performance and any sex-invariant MYH10 conclusion; it does not choose between conflicting statements.
